Este estudo aborda a variabilidade fitoquímica e a biossíntese de alcaloides medicinais em três espécies de "unha de gato": Uncaria rhynchophylla, U. tomentosa e U. guianensis. O problema central reside na variação da composição de alcaloides nessas plantas, influenciada por fatores ambientais e genéticos, o que impacta sua padronização para uso farmacêutico. Diante disso, o objetivo foi identificar genes e vias metabólicas envolvidos na biossíntese de alcaloides, além de apontar os fatores externos que afetam essa produção. Para alcançar os objetivos, foi realizada uma revisão sistemática da literatura, seguindo o protocolo PRISMA. A pesquisa foi conduzida nas bases de dados PubMed e Portal de Periódicos da CAPES, utilizando palavras-chave e operadores booleanos. Os critérios de inclusão e exclusão foram aplicados rigorosamente, resultando na seleção de 28 artigos originais para análise qualitativa. Os resultados revelaram que a U. rhynchophylla se destaca pela produção de alcaloides indólicos terpênicos, enquanto U. tomentosa e U. guianensis são ricas em alcaloides oxindólicos. A pesquisa também demonstrou que a produção desses compostos é sensível a fatores como estresse hídrico e luminosidade, e que a regulação molecular envolve genes e fatores de transcrição como UrPIF3 e WRKY. A principal conclusão é que, embora haja avanços, existem lacunas significativas, como a falta de dados genéticos sobre as espécies neotropicais e a escassez de estudos que correlacionem a variabilidade fitoquímica com as propriedades medicinais, destacando a necessidade de futuras pesquisas para otimizar a produção de alcaloides de interesse farmacêutico.
