Therefore, high metabolism and cellular differentiation is expectable, with consequent production of reactive oxygen species (ROS). These products of metabolism are known to cause oxidative stress, with detrimental effect on growth and survival of fish larvae. These organisms have effective antioxidant defences to protect cells from oxidative stress, but the efficiency of these mechanisms depends on the adequate supply of dietary antioxidants, such as vitamins. So, it is possible and evident that diet might play a protective role against ROS. The objective of this study was to assess the activity of enzymes of oxidative stress in the sea bass juveniles fed diet with different levels of vitamins. Sea bass post-larvae were reared between 30 to 70 days according to EPPO standard rearing procedures for this species. Three experimental diets A, B and C were tested in triplicate. Diets were formulated and produced by SPAROS. Growth and survival were monitored along the trial. At the end of the trial 30 post-larvae were collected per tank. Tissues were homogenized in phosphate buffered saline solution at pH 7.4 (1g of tissue/15ml buffer), centrifuged at 13,500g, at 4ºC, for 15 min. Supernatant was divided in different aliquots and stored at -80ºC until analysis. Aliquots were used for the determination of several parameters. Oxidative damage due to lipid peroxidation was evaluated by the formation of malondialdehyde (MDA). Superoxide dismutase (SOD) activity was determined using nitroblue tetrazolium (NBT) as substrate and the reaction was monitored at 560 nm. Glutathione peroxidase (GPx) activity was assayed using cumene hydroperoxide and hydrogen peroxide as substrates for measuring total GPX and Se-dependent GPX (Se-GPX), respectively, and the reaction monitored at 340 nm. Catalase (CAT) activity was determined using hydrogen peroxide as substrate and monitoring the reaction at 240nm. The activity of enzymes was normalized by the total protein content, being expressed as U/mg of protein. The results concerning the activity of oxidative stress enzymes of sea bass fed diets with different levels of vitamins will be presented in this study.
