地下水の流動は「トレーサー試験」によって把握するのが一般的である.本研究ではDNAを利用した新たなトレーサーを開発することを目的として,DNAトレーサーの作製とその有効性に関して基礎的な実験を行った.水中などの環境中のDNA分析ではフィルタリングしたものからDNAを抽出して,リアルタイム定量PCR分析をかけるため,むき出しのプラスミドDNAはトレーサーとして利用できないことがわかった.しかし,DNAをリポソームに封入した場合にはフィルターでのトラップが容易となり,DNAが96時間程度分解されないことが明らかとなった.また,このリポソーム封入DNAを用いて,1mの珪砂入り管路での地下水浸透実験を実施したところ,塩分とほぼ同様の動態を示し,地下水動態のトレーサーとして利用可能であることが示された.
