Étude des métabolites liés à l’îlot pks chez les souches urinaires de Escherichia coli Article Swipe
Escherichia coli is the first cause of symptomatic urinary tract infections but can also be involved in asymptomatic bacteriuria. The genome of some urinary strains of E. coli harbors a pathogenicity island, named pks island. Transcriptomic studies show its expression in urine, without any known role. It allows the biosynthesis of different metabolites : a genotoxin, the colibactin ; aminolipids, with activity on pain ; iron chelators, required for bacterial growth in urine. Before predicating a pathophysiological role of pks island in urinary strains, the aim of this work is to study its functionality in these strains of E. coli, by monitoring metabolites production by direct or indirect mean. The pks island is associated with genotoxicity with DNA double-strand breaks and DNA cross-link in vitro. Aminolipids are also produced thanks to this island in bacterial cultures. The pks island is therefore functional, either in E. coli ABU83972, an asymptomatic bacteriuria strain ; or in E. coli CFT073, an uropathogenic strain. However, the concentrations of metabolites differ depending on culture conditions and on strains studied, without a clear correlation with pathogenicity. This in vitro study has been completed by the identification and quantification of metabolites in urines of patients suffering from E. coli pyelonephritis. Aminolipids found in urine evidence for the first time the in vivo production of colibactin during an urinary tract infection.
