Evaluation of Antimicrobial Resistance and Characterization of Virulence Factors of Pseudomonas aeruginosa Isolates with Special Reference to Biofilm Formation Article Swipe

Background: Emergence of multi-drug resistant Pseudomonas aeruginosa (PA), coupled with the pathogen’s versatile virulence factors, leads to high morbidity and mortality rates. PA possesses a variety of virulence factors and infections caused by multidrug-resistant PA in clinical isolates are a public health problem. The aim of this study is to evaluate the antimicrobial susceptibility pattern of PA clinical isolates. Methods: A total of 3120 nonrepetitive clinical samples were collected from the Department of Microbiology in the tertiary care hospital, Government Thoothukudi Medical College, Thoothukudi, between August and September of 2022. Antibiotic susceptibility test was performed through the agar disk diffusion method. The ability of producing biofilm was examined by microtiter colorimetric biofilm assay (tissue culture plate method [TCPM]) and Congo red agar method, and the prevalence of the algD, PslD, and PelF genes among the isolates was determined by polymerase chain reaction. Results: Out of 52 isolates, 67.3% were susceptible PA (S-PA) and 32.69% were multidrug-resistant PA (MDR-PA). The most susceptible antibiotic was found to be aztreonam (90.3%), followed by meropenem (84.6%) whereas most of the organisms were found to be resistant to ciprofloxacin (32.6%), followed by levofloxacin (30.75%). In our study, 84% of the isolates were biofilm producers when detected phenotypically by TCPM, out of which 50% were strong, 26.92% were moderate, and 7.62% were weak biofilm producers. The prevalence of the AlgD, PslD, and PelF genes was 75%, 11.53%, and 1.92% among the isolates, respectively. Conclusions: The high rate of S-PA and MDR-PA in its ability to produce biofilm is an alarm for public health. The statistical analysis showed that biofilm production in the S-PA and MDR-PA isolates was statistically significant ( P < 0.03).
