Farmacocinética de cefquinome en terneros por aplicación intramuscular Article Swipe

Resumen Se determinaron parámetros farmacocinéficos de cefquinome referidos a su absorción, distribución y eliminación, tras la aplicación intramuscular de 1 mg/kg a terneros de 34 ± 25 días de edad. Los niveles plasmáticos se cuantificaron mediante método microbiológico utilizando medio de cultivo Antibiotic Nº 1 (Merck) y Providencia alcalifaciens como microorganismo detector y mediante la preparación de nueve diluciones estándares (25 a 0.097 µg/ml). Se confrontaron los diámetros de los halos de inhibición de las muestras versus la dilución estándar, utilizando una curva de calibración obtenida por regresión semilogarítmica (r2= 0.989). El límite de cuantificación del ensayo fue de 0.097 µg/ml. Los datos de concentración de cefquinome en función del tiempo fueron analizados con el software RESID e interpretados por el modelo bicompartimental (r2 = 0.9). Los resultados obtenidos son concordantes con el perfil cinético, exhibido por las cefalosporinas en los animales domésticos y lo informado para cefquinome en bovinos adultos e indican que la aplicación intramuscular produce rápida absorción y distribución, generando niveles plasmáticos máximos a las 0.85 ± 0.11 horas post aplicación, mientras que la difusión es restringida y breve la permanencia en el organismo, no obstante se comprobó niveles plasmáticos superiores a los terapéuticos que subsisten durante 6 horas post administración. Según los parámetros, farmacocinéticos obtenidos se estableció una dosis de mantenimiento de 1. 22 mg/kg de peso aplicados cada 24 horas por via intramuscular. Summary Pharmacokinetic parameters of cefquinome referred to their absorption and elimination, were determined after a single intramuscular dose 1 mg/kg to calves 34 ± 25 days old. The plasmatic levels were quantified by microbiological assay in agar Antibiotic Nº 1 (Merck) using Providencia alcalifaciens as test microorganism by means of the preparation of nine standard dilutions (25 at 0.097 µg/ml). The diameters of inhibition zones of the samples were confronted versus the standard dilution, using a calibration curve obtained by semilogaritmic regression (r2= 0.989). The quantification limit was of 0.097 (µg/ml). The data of cefquinome concentration in function of the time were analyzed with the software RESID and interpreted as a bicompartimental model (r2= 0.9). The obtained results are concordant with the kinetic profile exhibited by cephalosporins in domestic animals and that informed for cefquinome in bovine adults and they indicate that the intramuscular application produces quick absorption and distribution, generating maximun plasmatic levels at 0.85 ± 0.11 hours post application, while the diffusion is restricted and brief the permanency in the organism, nevertheless superior plasmatic levels to the therapeutic ones was proven, that subsist during 6 hours post administration. According to the pharmacokinefic parameters obtained, a dose of maintenance of 1.22 mg/kg was settled down every 24 hours. Key words: bovines, cephalosporins, disposition.
