Gram-scale synthesis of calix[n]furans (n = 4, 5, and 6) has been achieved using size-selective precipitation protocols. The obtained calix[n]furans were successfully converted to cyclic polyketones and subsequently to ethylene-bridged cyclic isopyrazoles in moderate yields. Single crystal X-ray diffraction analysis revealed the solid-state conformations of these macrocycles. The cyclic isopyrazoles formed well-defined cavities with square, pentagonal, and rectangular shapes for n = 4, 5, and 6, respectively. Cyclic penta- and hexaisopyrazoles were found to crystallize with solvent molecules encapsulated within the cavity, suggesting their rich host-guest chemistry.
