Improving the understanding of mammalian microRNAs: from annotation, isomiR and function Article Swipe
MicroRNAs (miRNAs) are a class of small non-coding RNAs, ~22nt. They have extensive roles in gene expression regulation, fine-tuning a wide range of biological processes. Next-generation sequencing is currently a widely used approach to study miRNA expression level in samples. A bioinformatic analysis pipeline commonly implemented to investigate the miRNA-seq data. \nWe find that (1) adapter information is important in the small RNA-seq studies; (2) miRNA annotation content in miRBase need to be curtated before used in mapping as reference; (3) developed a hierarchical isomiR nomenclature and implement it within a publicly available analysis pipeline, Jasmine; (4) developed a deep neural network based approach, miRAW, for miRNA target prediction.\nOverall, the works presented in this thesis demonstrate that bioinformatic analysis pipeline can affect our understanding of mammalian microRNAs, to some degree; and highlight the importance of implementing robust and reliable bioinformatic methods in miRNA-seq studies, especially in medical related research fields.
