In vivo vaccine generation with a topical small molecule cocktail to eradicates AML and PDAC Article Swipe

Using vaccine to cure cancer has been expected for over 130 years, but remains a big challenge. Here we present a novel in vivo personal and off-the-shelf vaccine generation strategy with unique mechanism, which is very simple but high efficacy, using a small molecule cocktail as antigen-agnostic vaccine. In mouse AML and PDAC models, this in vivo vaccine rapidly eliminated tumors via triggering inflamed immunogenic cell death to release antigens, activating NK and DC cells, inducing innate and adaptive immunity in blood, lymph node, spleen, bone marrow and TME, and elicited a robust long-term immune memory. Importantly, this small molecule cocktail-based platform holds the potential to make in vivo vaccine more personal and accessible. It might represent a promising avenue for pan-cancer immunotherapy.
