Insulin is a peptide hormone secreted from pancreatic β cells to regulate blood glucose levels. Insulin maturation is dysregulated in diabetes and there are many open questions on how the cell regulates maturation. Maturation of insulin involves acidification of the vesicle lumen and cleavage of proinsulin to insulin. This cleavage step allows for insulin to condense and crystallize within the vesicle for storage prior to secretion. We have developed two approaches for mapping and quantifying insulin maturation within cells. We use soft X-ray tomography to map organelle organization and determine the molecular density of secretory vesicles which is not possible with other methods. Additionally, we developed a novel fluorescence lifetime imaging microscopy (FLIM) pH probe to monitor vesicle pH in live cells. Using these approaches with various chemical probes allows us to explore how different protein signaling networks impact insulin maturation over time. We have also developed a novel visual design language for representing multi-scale structural data for sharing our results with lay audiences to teach basic concepts of structural biology and diabetes.
