Distinct oxytocin signaling pathways synergistically mediate rescue-like behavior in mice
2025-04-23 • Fengrui Zhang, Juan Liu, Jieqi Wen, Zi-Yan Zhang, Yijia Li, Eric Song, Li Hu, Zhou‐Feng Chen
Spontaneous rescue behavior enhances the well-being and survival of social animals, yet the neural mechanisms underlying the recognition and response to conspecifics in need remain unclear. Here, we report that observer mice experience distress when encountering anesthetized conspecifics, prompting spontaneous rescue-like behavior toward the unconscious mice. This behavior facilitates the earlier awakening of anesthetized mice while simultaneously alleviating stress in the helper mice. Our findings reveal that end…

Structures of human gastrin-releasing peptide receptors bound to antagonist and agonist for cancer and itch therapy
2023-02-01 • Shuman Peng, Yuting Zhan, Dongqi Zhang, Lu Ren, Anqi Chen, Zhou‐Feng Chen, Haitao Zhang
Gastrin releasing peptide receptor (GRPR), a member of the bombesin (BBN) G protein-coupled receptors, is aberrantly overexpressed in several malignant tumors, including those of the breast, prostate, pancreas, lung, and central nervous system. Additionally, it also mediates non-histaminergic itch and pathological itch conditions in mice. Thus, GRPR could be an attractive target for cancer and itch therapy. Here, we report the inactive state crystal structure of human GRPR in complex with the non-peptide antagonis…

Molecular and neural basis of pleasant touch sensation
2022-04-28 • Benlong Liu, Lina Qiao, Kun Liu, Juan Liu, Tyler J. Piccinni-Ash, Zhou‐Feng Chen
Pleasant touch provides emotional and psychological support that helps mitigate social isolation and stress. However, the underlying mechanisms remain poorly understood. Using a pleasant touch–conditioned place preference (PT-CPP) test, we show that genetic ablation of spinal excitatory interneurons expressing prokineticin receptor 2 (PROKR2), or its ligand PROK2 in sensory neurons, abolishes PT-CPP without impairing pain and itch behaviors in mice. Mutant mice display profound impairments in stress response and p…

BNP facilitates NMB-encoded histaminergic itch via NPRC-NMBR crosstalk
2021-12-17 • Qingtao Meng, Xian‐Yu Liu, Xueting Liu, Juan Liu, Admire Munanairi, Devin M. Barry, Benlong Liu, Hua Jin, Yu Sun, Qianyi Yang, Fang Gao, Li Wan, Ji...
Histamine-dependent and -independent itch is conveyed by parallel peripheral neural pathways that express gastrin-releasing peptide (GRP) and neuromedin B (NMB), respectively, to the spinal cord of mice. B-type natriuretic peptide (BNP) has been proposed to transmit both types of itch via its receptor NPRA encoded by Npr1 . However, BNP also binds to its cognate receptor, NPRC encoded by Npr3 with equal potency. Moreover, natriuretic peptides (NP) signal through the G i -couped inhibitory cGMP pathway that is supp…

GRP and NMB: distinct functions in itch transmission
2021-01-01 • Devin M. Barry, Xian‐Yu Liu, Zhou‐Feng Chen
One of the major challenges in the field is to understand how histaminergic and nonhistaminergic itch is encoded and transmitted from sensory neurons to the spinal cord, respectively. The question is of immerse importance because it concerns not only the coding of itch information in the nervous system but also antihistamines are ineffective at treating the majority of chronic itch. One way to tackle the challenge is to elucidate the role of neuropeptides and their respective receptors in itch transmission. This t…
